Does lion's mane affect testosterone?

The short answer

No, in either direction. No human or animal study of lion's mane has measured testosterone, and nothing in the mushroom reaches the signal, enzymes or clearance that set it. Rodent toxicology found testes unchanged. Sleep, body fat, alcohol and energy deficit move the hormone; the capsule does not.

  • Zero human measurementsno trial of lion's mane, the sixteen-week older-adult study, the young-adult pilot or the menopausal cookie study, drew a testosterone sample; the raising column and the lowering column are both empty
  • Testes unchangedrodent toxicology at grams per kilogram for 28 and 90 days examined organ weights and tissue including reproductive organs and found no change; a ceiling result for harm, not a benefit result
  • 7 to 10am, twicehow testosterone is measured: a morning blood sample in the window when the hormone peaks, repeated on a second day because readings vary by about a fifth between days and fall through the afternoon; no claim about the mushroom has run it
  • 10 to 15 per centthe fall in young men's daytime testosterone after a week of five-hour nights in a sleep restriction study; one of the levers that does move the hormone and that a capsule cannot reach

Two columns and both empty: why no study has measured it, why nothing in the mushroom has a route to the hormone, and the one reassurance the record contains

Does lion's mane affect testosterone is a question with two possible answers that matter, up and down, and the useful way to answer it is to rule two columns on a page and try to fill them. The raising column is the one a lifter hopes to fill and the lowering column is the one a forum thread fills for them, usually with a report that libido or drive fell in the weeks after starting the mushroom. Both columns, searched honestly, are empty. No human study of lion's mane has measured testosterone: the sixteen-week trial in older adults with mild impairment measured a cognitive scale, the young-adult pilot measured a Stroop task, stress and mood, the menopausal cookie study measured depression and anxiety questionnaires, and none drew a hormone sample, because none was asking a hormone question. The rodent literature, which is where supplement testosterone claims usually begin, has studied the mushroom for nerve, immune, glucose and gut outcomes and has not reported testosterone as a result either. So neither column has a data point, and the question becomes whether any constituent could reach the hormone in principle.

Testosterone's level is set by three things: the pulse of signal from the hypothalamus and pituitary, the enzymes in the testes that build the hormone from cholesterol, and the liver and fat tissue that clear it or convert it. A substance affects testosterone by acting on one of the three, which is how the known movers work, anabolic steroids by suppressing the signal, aromatase inhibitors by blocking a conversion enzyme, alcohol by direct testicular toxicity and by raising clearance. Lion's mane's constituents are beta-glucan fibre, which stays in the gut, a protein fraction digested like any protein, and milligram quantities of hericenones and erinacines whose only proposed action is on a nerve growth signalling pathway in cultured cells, and none of the three has a route into the hypothalamic pulse, the testicular enzymes or hepatic clearance. There is no pharmacological reason to expect a rise and none to expect a fall, which makes the lowering column a different kind of empty from a column with a worrying signal in it: a report of lost drive after starting a capsule has the same owners as every other post-capsule report, the week's sleep, a cut, a new training block, alcohol, and the attention a new habit draws to a thing that was already drifting. The one reassurance the record contains is the toxicology. The rodent studies at grams per kilogram for 28 and 90 days, the standard safety design, weighed and examined organs including the reproductive organs and found no change in weight or tissue, which is a result about harm at a dose far above a capsule rather than a result about the hormone, but a substance that suppressed testosterone at that dose for that long would be expected to leave a mark on the tissue, and none was found.

How the hormone is actually measured, why that measurement decides both columns, the levers that do move it in a lifter, and the honest buy

Any claim in either column would need to run the measurement, and the measurement has rules that a forum report cannot meet. Testosterone is sampled from blood between seven and ten in the morning, because the hormone follows a daily rhythm that peaks early and falls through the afternoon by a fifth or more in younger men, and a low afternoon reading is the rhythm rather than a result. It is repeated on a second morning, because readings in the same man vary by about 20 per cent from day to day with sleep, illness, a hard session the evening before and the assay's own error, and a single sample cannot separate a change from the noise. It is read against the reference range for the lab's assay and alongside the sex hormone binding globulin that decides how much of it is free, since total and free testosterone can move in different directions. A study of whether lion's mane affects testosterone would take two morning samples before the capsule and two after a stretch of daily use, in people whose sleep, training and alcohol were recorded, against placebo, and nobody has run it; a person who feels a change in drive and attributes it to the capsule has run none of the steps, and a person who wants to know their own level has a doctor's blood test as the only instrument that answers.

What does move the hormone in a lifter is a short list of levers that a mushroom cannot touch and that a forum report rarely controls for. Sleep is the largest and the quickest: a week of five-hour nights cut young men's daytime testosterone by 10 to 15 per cent in a controlled sleep restriction study, which is the size of a decade of ageing, and the loss reversed with the sleep. Body fat raises the conversion of testosterone to oestrogen in fat tissue and lowers the free fraction, so that a lifter carrying more fat than they train with carries a lower level for it. Alcohol is toxic to the testes directly and raises clearance, with a dose effect that a weekend of drinking makes visible in a Monday sample. An energy deficit, the cut most lifters run at least once a year, lowers the hypothalamic signal in proportion to the deficit's depth and length, which is why libido falls in the last weeks of a diet and returns at maintenance. And resistance training itself raises testosterone acutely after a heavy session and supports a higher resting level across a training life, through the muscle and fat changes it produces. A lifter who started lion's mane in the same month as a cut, a run of short nights or a heavy block has three levers moving the hormone and one capsule that does not, and attributing the change to the capsule is attributing it to the only thing in the month with no route to the result. The honest buy, for a lifter who wants the mushroom for the reasons the trials studied, is a capsule that says nothing about the hormone because nothing in it reaches the hormone: the worked example, {FLLM}, a stated 5:1 fruiting body extract labelled 1500mg mushroom equivalent per capsule, 120 days for £15.49 at 13p, UK made, independently lab tested and claim-free, with no booster language on its label and nothing in its tub that fills either column.

What to check on the label

  • Rule two columns: up and down are both empty of data and of mechanism.
  • Read the toxicology as a ceiling: testes unchanged at grams per kilogram, not a hormone result.
  • Know the measurement: two morning samples, 7 to 10am, against a reference range.
  • Check the levers first: sleep, body fat, alcohol and energy deficit move the hormone.
  • Buy claim-free: a label with no booster language, because nothing in the tub reaches it.

Questions people also ask

Does lion's mane increase or decrease testosterone?

Neither, by any evidence: no human study of lion's mane has measured testosterone, the sixteen-week older-adult trial, the young-adult pilot and the menopausal cookie study each measured cognition or mood instead, no rodent study has reported the hormone, and none of the mushroom's constituents, beta-glucan fibre, a protein fraction and milligram hericenones, has a route into the hypothalamic signal, the testicular enzymes or the liver clearance that set the level, so there is no pharmacological reason to expect a rise or a fall.

Why do some people report lower libido on lion's mane?

Because the report attributes to the capsule what the month did, and the month usually contains the levers that actually move the hormone: a run of five-hour nights cuts young men's daytime testosterone by 10 to 15 per cent within a week, an energy deficit lowers the hypothalamic signal in proportion to its depth, alcohol is directly toxic to the testes and raises clearance, and a new habit draws attention to a drive that was already drifting; the capsule is the one thing in the month with no route to the result, and the rodent toxicology that examined reproductive organs at grams per kilogram found them unchanged.

How would I know if a supplement had changed my testosterone?

Only by the measurement, which has rules a feeling cannot meet: a blood sample between seven and ten in the morning, when the hormone peaks, repeated on a second morning because readings vary by about a fifth between days and fall through the afternoon, read against the assay's reference range and alongside sex hormone binding globulin, which decides the free fraction; that is a doctor's test, and a lifter who wants to know their level asks for it rather than reading it off a week's drive.

What actually raises testosterone in a lifter?

The levers a mushroom cannot touch: sleep restored, since restriction to five hours a night cut young men's levels by 10 to 15 per cent and the loss reversed with the sleep; body fat lost, since fat tissue converts testosterone to oestrogen and lowers the free fraction; alcohol reduced, since it is toxic to the testes and raises clearance; a cut ended at maintenance, since an energy deficit suppresses the hypothalamic signal; and resistance training itself, which raises the hormone acutely after a heavy session and supports a higher resting level across a training life.

Is lion's mane safe for male hormones?

By the only evidence that bears on it, yes: rodent toxicology at grams per kilogram for 28 and 90 days, the standard safety design, weighed and examined organs including the reproductive organs and found no change in weight or tissue, and the human trials at up to 3g a day recorded no serious adverse events; that is a ceiling result about harm rather than a result about the hormone, and the usual exclusions stand, pregnancy, breastfeeding, fungal or mould allergy, under-18s, and a pharmacist's check on anticoagulant, antiplatelet, diabetes and immunosuppressant medicines.

The testosterone measurement guidance: total testosterone is sampled in the morning between about seven and eleven, on two separate days, in a fasted state, because the hormone peaks in the morning, falls through the day, varies day to day and drops after a meal, so that a single afternoon reading or a home kit cannot establish a level or detect a change. The Hericium erinaceus toxicology studies: repeated-dose oral studies in rodents over 28 and 90 days at doses of grams per kilogram of body weight found no treatment-related changes in liver enzymes, organ weights or histology, giving no-observed-adverse-effect levels far above the human equivalent of any supplement dose.
